I think you'll find that is one of those quotes attributed to a certain movie star, in this case John Wayne, that was never actually said in a film. Just as Cary Grant never said "Judy, Judy, Judy" and James Cagney didn't exactly say "You dirty rat."

But here is a nice story about an actual John Wayne movie quote. He was one of the many stars in the biblical film King Of Kings and was wildly miscast to play the centurion at Christ's crucifixion (he also once played Genghis Khan, but that's another story). He had only one line: "Truly this is the son of God" which he spoke the same way he might say "That's a mighty nice horse you got there, pardner." After several takes, the director took him aside and suggested that he needed to inject more awe into the reading. Wayne nodded and for the next take said: "Aw, truly this is the son of God."

To revive the movie quote competition, how about this one:

"You're gonna need a bigger boat."

I believe the quote is correct. It was said by Roy Scheider (to Robert Shaw). Both Scheider and Richard Dreyfuss wore glasses in the film.
